Sertaconazole In-Situ Gel for Vulvovaginal Candidiasis

Introduction: The rationale of this investigation is to formulate thermosensitive mucoadhesive in situ gel of sertaconazole for management of vulvovaginal candidiasis, with the aim to prolong residence time in vagina, control drug release, and enhance efficacy as well as improvement in patient compliance. In situ gel formulations of sertaconazole were formulated using different concentration of pluronic-127 alone and in combination with pluronic-68 as thermosensitive polymer and HPMC K4M also added as mucoadheive polymer. The in situ gelling formulations were characterized for their gelation temperature, gel melting temperature, mucoadhesive force, microbiological studies and in vitro release studies. It was found that the viscosity and mucoadhesive force increased as the concentration of pluronic-127 increased and in vitro drug release decreased. In conclusion, HPMC K4M was capable of enhancing the sustain release property of pluronics demonstrating that the blend of HPMC K4M and pluronics possibly will exploit in formulation of vaginal in situ gel drug delivery system with prolong local residence time and as a result superior clinical effect.
